Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_021939.4(FKBP10):c.1399+1G>A, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site results in skipping of exon 8 and activation of a cryptic splice donor site and introduces a premature termination codon (PMID: 22061863). The resulting mRNA is expected to undergo nonsense-mediated decay. Disruption of this splice site has been observed in individual(s) with osteogenesis imperfecta (PMID: 22061863). This variant is present in population databases (no rsID available, gnomAD no frequency). This sequence change affects a donor splice site in intron 8 of the FKBP10 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product.
