Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001287.6(CLCN7):c.637C>T (p.Leu213Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 213 of the CLCN7 protein (p.Leu213Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant osteopetrosis (PMID: 12929941, 16120485, 26395888). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CLCN7 protein function. For these reasons, this variant has been classified as Pathogenic.
