This variant lies in the CAPN3 gene (transcript NM_000070.3) at coding-DNA position 413, where T is replaced by C; at the protein level this means replaces leucine at residue 138 with proline — a missense variant. Submitter rationale: This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 138 of the CAPN3 protein (p.Leu138Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive limb-girdle muscular dystrophy (PMID: 17562833). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals.
