Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251496 control chromosomes (gnomAD). c.1454G>T has been observed in individuals affected with Oculocutaneous Albinism (Rooryck_2008, Grnskov_2008, Lasseaux_2018). These data indicate that the variant may be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.1453G>A, p.Gly485Arg), supporting the critical relevance of codon 485 to OCA2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
