Pathogenic for DNA ligase IV deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_206937.2(LIG4):c.839A>G (p.Gln280Arg), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt LIG4 protein function. This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 280 of the LIG4 protein (p.Gln280Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with severe combined immunodeficiency (PMID: 16358361). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals.
