NM_001370259.2(MEN1):c.1150G>T (p.Glu384Ter) was classified as Pathogenic for Multiple endocrine neoplasia, type 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This premature translational stop signal has been observed in individual(s) with multiple endocrine neoplasia type 1 (PMID: 25309785). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu384*) in the MEN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MEN1 are known to be pathogenic (PMID: 12112656, 17853334).
