This variant lies in the FGFR2 gene (transcript NM_000141.5) at coding-DNA position 1084, where G is replaced by T; at the protein level this means replaces alanine at residue 362 with serine — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 362 of the FGFR2 protein (p.Ala362Ser). This variant also falls at the last nucleotide of exon 8, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with Crouzon syndrome (PMID: 10574673; internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 2735493). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
