This variant lies in the KIF11 gene (transcript NM_004523.4) at coding-DNA position 730, where C is replaced by T; at the protein level this means replaces histidine at residue 244 with tyrosine — a missense variant. Submitter rationale: This sequence change replaces histidine, which is basic and polar, with tyrosine, which is neutral and polar, at codon 244 of the KIF11 protein (p.His244Tyr). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with congenital microcephaly and chorioretinopathy with or without lymphedema (PMID: 25115524, 26566857). In at least one individual the variant was observed to be de novo.
