This variant lies in the RET gene (transcript NM_020975.6) at coding-DNA position 1763, where G is replaced by A; at the protein level this means replaces glycine at residue 588 with aspartic acid — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This missense change has been observed in individual(s) with Hirschsprung disease (PMID: 10946353). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 588 of the RET protein (p.Gly588Asp).
