Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000497.4(CYP11B1):c.1322T>G (p.Val441Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 441 of the CYP11B1 protein (p.Val441Gly). This variant is present in population databases (rs772169059, gnomAD 0.0009%). This missense change has been observed in individual(s) with 11-beta-hydroxylase deficiency (PMID: 8506298, 28228528; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 2735225). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CYP11B1 protein function. Experimental studies have shown that this missense change affects CYP11B1 function (PMID: 8506298, 20089618). For these reasons, this variant has been classified as Pathogenic.
