Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 4e-05 in 251398 control chromosomes. This frequency is not significantly higher than estimated for disease-causing variants in DDC, allowing no conclusion about variant significance. c.1340G>A has been observed in individuals affected with Deficiency Of Aromatic-L-Amino-Acid Decarboxylase (e.g. Verbeek_2007, Spitz_2017, Gorukmez_2023, Himmelreich_2023). These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.1339C>T, p.Arg447Cys), supporting the critical relevance of codon 447 to DDC protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
