Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198239.2(CCN6):c.866dup (p.Ser290fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with autosomal recessive progressive pseudorheumatoid dysplasia (PMID: 21993478, 22685593). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs782450077, gnomAD 0.006%). This sequence change creates a premature translational stop signal (p.Ser290Glufs*13) in the WISP3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 65 amino acid(s) of the WISP3 protein.
