Pathogenic for Methylmalonic acidemia — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000255.4(MMUT):c.599T>C (p.Ile200Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MUT c.599T>C (p.Ile200Thr) results in a non-conservative amino acid change located in the methylmalonyl-CoA mutase N-terminal domain (IPR006098) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251274 control chromosomes. c.599T>C has been reported in the literature in multiple homozygous and compound heterozygous individuals affected with Methylmalonic Acidemia (Han_2015, Kang_2020, Yu_2021) of in a heterozygous individual with Methylmalonic Acidemia without detection of a second allele change (Liu_2012). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
