Disruption of the initiator codon has been observed in individual(s) with clinical features of Treacher-Collins syndrome (PMID: 15340364, 22317976, 25790162). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change affects the initiator methionine of the TCOF1 mRNA. The next in-frame methionine is located at codon 121.
