Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000539.3(RHO):c.937-1G>T, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the RHO gene (transcript NM_000539.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 937, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 4 of the RHO gene. While this variant is not anticipated to result in nonsense mediated decay, it likely alters RNA splicing and results in a disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. Disruption of this splice site has been observed in individuals with clinical features of autosomal dominant retinitis pigmentosa (PMID: 28045043). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency).
