This variant lies in the CYP1B1 gene (transcript NM_000104.4) at coding-DNA position 1133, where T is replaced by A; at the protein level this means replaces leucine at residue 378 with glutamine — a missense variant. Submitter rationale: This sequence change replaces leucine, which is neutral and non-polar, with glutamine, which is neutral and polar, at codon 378 of the CYP1B1 protein (p.Leu378Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with primary congenital glaucoma (PMID: 19528825, 27268095). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
