Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001206744.2(TPO):c.2266T>C (p.Cys756Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 756 of the TPO protein (p.Cys756Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive thryoid dyshormonogenesis (PMID: 23512414, 26174974). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. For these reasons, this variant has been classified as Pathogenic.
