This variant lies in the CDC73 gene (transcript NM_024529.5) at the canonical splice donor site of the intron immediately after coding-DNA position 307, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 3 of the CDC73 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with CDC73-related conditions (PMID: 23029104, 24716902, 32590342). This variant is also known as IVS3+1 G>A.
