Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001254.4(CDC6):c.1253A>G (p.Lys418Arg), citing Invitae Variant Classification Sherloc (09022015): An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with CDC6-related conditions. This variant is present in population databases (rs779176597, gnomAD 0.004%). This sequence change replaces lysine, which is basic and polar, with arginine, which is basic and polar, at codon 418 of the CDC6 protein (p.Lys418Arg). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
