Uncertain significance for Tuberous sclerosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000548.5(TSC2):c.-30+2_-30+22del, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TSC2 gene (transcript NM_000548.5) at the canonical splice donor site of the intron immediately after 30 bases upstream of the translation start (5' untranslated region) through 22 bases into the intron immediately after 30 bases upstream of the translation start (5' untranslated region), deleting this region. Submitter rationale: This sequence change falls in intron 1 of the TSC2 gene. It does not directly change the encoded amino acid sequence of the TSC2 protein. It affects a nucleotide within the consensus splice site. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TSC2-related conditions. ClinVar contains an entry for this variant (Variation ID: 2730611). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
