Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.4141A>G (p.Lys1381Glu), citing Ambry Variant Classification Scheme 2023: The p.K1381E variant (also known as c.4141A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 4141. The lysine at codon 1381 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
