Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001256715.2(DNAAF3):c.40G>C (p.Val14Leu), citing Ambry Variant Classification Scheme 2023: The c.181G>C (p.V61L) alteration is located in exon 2 (coding exon 2) of the DNAAF3 gene. This alteration results from a G to C substitution at nucleotide position 181, causing the valine (V) at amino acid position 61 to be replaced by a leucine (L). Based on data from gnomAD, the C allele has an overall frequency of 0.002% (5/247594) total alleles studied. The highest observed frequency was 0.028% (5/17964) of East Asian alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
