Pathogenic for Combined deficiency of sialidase AND beta galactosidase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000308.4(CTSA):c.518del (p.Phe173fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Phe191Serfs*2) in the CTSA gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CTSA are known to be pathogenic (PMID: 15110321, 23915561). This variant is present in population databases (rs769812697, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with CTSA-related conditions.
