Pathogenic for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001277115.2(DNAH11):c.8407_8416del (p.Asp2803fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DNAH11 gene (transcript NM_001277115.2) at coding-DNA position 8407 through coding-DNA position 8416, deleting 10 bases; at the protein level this means shifts the reading frame starting at aspartic acid residue 2803, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has not been reported in the literature in individuals affected with DNAH11-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asp2803Cysfs*2) in the DNAH11 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DNAH11 are known to be pathogenic (PMID: 18022865, 20513915, 22184204).
