Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001903.5(CTNNA1):c.2515G>A (p.Ala839Thr), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CTNNA1 protein function. This variant has not been reported in the literature in individuals affected with CTNNA1-related conditions. This variant is present in population databases (no rsID available, gnomAD 0.007%). This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 839 of the CTNNA1 protein (p.Ala839Thr). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
