NM_020750.3(XPO5):c.625A>T (p.Thr209Ser) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with XPO5-related conditions. This variant is present in population databases (rs201099159, gnomAD 0.009%). This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 209 of the XPO5 protein (p.Thr209Ser). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
