Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.906A>G (p.Ile302Met), citing Ambry Variant Classification Scheme 2023: The p.I302M variant (also known as c.906A>G), located in coding exon 7 of the DICER1 gene, results from an A to G substitution at nucleotide position 906. The isoleucine at codon 302 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
