Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.4123C>T (p.Arg1375Cys), citing Ambry Variant Classification Scheme 2023: The p.R1375C variant (also known as c.4123C>T), located in coding exon 23 of the DNAH11 gene, results from a C to T substitution at nucleotide position 4123. The arginine at codon 1375 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
