Uncertain significance for Cardiac anomalies - developmental delay - facial dysmorphism syndrome — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_015335.5(MED13L):c.1136G>A (p.Arg379Gln), citing ACMG Guidelines, 2015: The missense variant c.1136G>A (p.Arg379Gln) in the MED13L gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is reported with the allele frequency (0.001%) in the gnomAD Exomes. The amino acid Arg at position 379 is changed to a Gln changing protein sequence and it might alter its composition and physico-chemical properties. Computational evidence (Polyphen, SIFT and MutationTaster) predicts conflicting evidence on protein structure and function for this variant. The amino acid change p.Arg379Gln in MED13L is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
