NM_005585.5(SMAD6):c.295G>A (p.Gly99Ser) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G99S variant (also known as c.295G>A), located in coding exon 1 of the SMAD6 gene, results from a G to A substitution at nucleotide position 295. The glycine at codon 99 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
