Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.647A>G (p.Glu216Gly), citing Ambry Variant Classification Scheme 2023: The p.E216G variant (also known as c.647A>G) is located in coding exon 4 of the TGFB3 gene. The glutamic acid at codon 216 is replaced by glycine, an amino acid with similar properties. This change occurs in the first base pair of coding exon 4. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
