Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.3148A>C (p.Thr1050Pro), citing Ambry Variant Classification Scheme 2023: The p.T1050P variant (also known as c.3148A>C), located in coding exon 24 of the FBN2 gene, results from an A to C substitution at nucleotide position 3148. The threonine at codon 1050 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
