Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016929.5(CLIC5):c.209C>T (p.Thr70Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 229 of the CLIC5 protein (p.Thr229Met). This variant is present in population databases (rs200737296, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with CLIC5-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
