This variant lies in the COL6A1 gene (transcript NM_001848.3) at coding-DNA position 235, where C is replaced by T; at the protein level this means replaces arginine at residue 79 with cysteine — a missense variant. Submitter rationale: Variant summary: COL6A1 c.235C>T (p.Arg79Cys) results in a non-conservative amino acid change located in the von Willebrand factor, type A domain (IPR002035) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.3e-05 in 236844 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
