Likely pathogenic for Diamond-Blackfan anemia — the classification assigned by Ambry Genetics to NM_000969.5(RPL5):c.3+5G>C, citing Ambry Variant Classification Scheme 2023: The c.3+5G>C intronic alteration consists of a G to C substitution 5 nucleotides after exon 1 (coding exon 1) of the RPL5 gene. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. RNA studies have demonstrated that this alteration results in abnormal splicing in the set of samples tested (Ambry internal data). In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. Based on the available evidence, this alteration is classified as likely pathogenic.
