Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.4264T>A (p.Tyr1422Asn), citing Ambry Variant Classification Scheme 2023: The p.Y1401N variant (also known as c.4201T>A), located in coding exon 31 of the NF1 gene, results from a T to A substitution at nucleotide position 4201. The tyrosine at codon 1401 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
