NM_000165.5(GJA1):c.32T>A (p.Leu11His) was classified as Pathogenic for Oculodentodigital dysplasia, autosomal recessive by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine, which is neutral and non-polar, with histidine, which is basic and polar, at codon 11 of the GJA1 protein (p.Leu11His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of GJA1-related conditions. (Invitae). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt GJA1 protein function with a positive predictive value of 80%. This variant disrupts the p.Leu11 amino acid residue in GJA1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 16709485, 19638688, 28258662). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
