Pathogenic for Autoimmune lymphoproliferative syndrome due to CTLA4 haploinsufficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005214.5(CTLA4):c.415T>C (p.Tyr139His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with histidine, which is basic and polar, at codon 139 of the CTLA4 protein (p.Tyr139His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with CTLA4-related conditions (Invitae). In at least one individual the variant was observed to be de novo. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Tyr139 amino acid residue in CTLA4. Other variant(s) that disrupt this residue have been determined to be pathogenic (Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
