Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.676T>G (p.Cys226Gly), citing Ambry Variant Classification Scheme 2023: The p.C226G variant (also known as c.676T>G), located in coding exon 5 of the PTCH1 gene, results from a T to G substitution at nucleotide position 676. The cysteine at codon 226 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
