Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6378C>A (p.Asn2126Lys), citing Ambry Variant Classification Scheme 2023: The p.N2126K variant (also known as c.6378C>A), located in coding exon 46 of the POLE gene, results from a C to A substitution at nucleotide position 6378. The asparagine at codon 2126 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
