Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033641.4(COL4A6):c.4831G>C (p.Glu1611Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 1612 of the COL4A6 protein (p.Glu1612Gln). This variant is present in population databases (rs774819718, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with COL4A6-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt COL4A6 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
