Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002547.3(OPHN1):c.899dup (p.Met300fs), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with OPHN1-related conditions. For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Met300Ilefs*11) in the OPHN1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in OPHN1 are known to be pathogenic (PMID: 12807966).
