This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 2977, where C is replaced by T; at the protein level this means replaces histidine at residue 993 with tyrosine — a missense variant. Submitter rationale: The p.H993Y variant (also known as c.2977C>T), located in coding exon 19 of the ATM gene, results from a C to T substitution at nucleotide position 2977. The histidine at codon 993 is replaced by tyrosine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al.
