Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.206A>T (p.Asp69Val), citing Ambry Variant Classification Scheme 2023: The p.D69V variant (also known as c.206A>T), located in coding exon 2 of the RAD50 gene, results from an A to T substitution at nucleotide position 206. The aspartic acid at codon 69 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
