Likely pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016180.5(SLC45A2):c.2T>G (p.Met1Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change affects the initiator methionine of the SLC45A2 mRNA. The next in-frame methionine is located at codon 37. This variant is not present in population databases (gnomAD no frequency). Disruption of the initiator codon has been observed in individual(s) with oculocutaneous albinism (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
