Submitter rationale: ABCB4 c.2064+3A>T is an intronic variant located in the donor splice region of intron 16. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:37471416;30284531). The variant was found to segregate with disease in at least one affected family (PMID:30284531). It has been observed in trans with a pathogenic or likely pathogenic variant (PMID:37471416). At least one splicing study demonstrated this variant results in aberrant splicing (PMID:34906502;30284531). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify ABCB4 c.2064+3A>T as a likely pathogenic variant.
