Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000094.4(COL7A1):c.2622C>A (p.His874Gln), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 2622, where C is replaced by A; at the protein level this means replaces histidine at residue 874 with glutamine — a missense variant. Submitter rationale: Variant summary: COL7A1 c.2622C>A (p.His874Gln) results in a non-conservative amino acid change located in the Fibronectin type III domain (IPR003961) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 245452 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2622C>A in individuals affected with Dystrophic Epidermolysis Bullosa, Recessive and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
