Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000094.4(COL7A1):c.1334G>A (p.Arg445Gln), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 1334, where G is replaced by A; at the protein level this means replaces arginine at residue 445 with glutamine — a missense variant. Submitter rationale: Variant summary: COL7A1 c.1334G>A (p.Arg445Gln) results in a conservative amino acid change located in the Fibronectin type III domain (IPR003961) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251408 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1334G>A in individuals affected with Dystrophic Epidermolysis Bullosa, Recessive and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
