Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004970.3(IGFALS):c.1483dup (p.Glu495fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: IGFALS c.1483dupG (p.Glu495GlyfsX42) results in a premature termination codon, predicted to cause a truncation of the encoded protein, however nonsense mediated decay is not expected. No pathogenic variants were observed downstream. The variant allele was found at a frequency of 1.4e-05 in 218022 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1483dupG in individuals affected with Short Stature Due To Primary Acid-Labile Subunit Deficiency and no experimental evidence demonstrating its impact on protein function have been reported.
