Submitter rationale: Variant summary: CFTR c.2519T>C (p.Ile840Thr) results in a non-conservative amino acid change located in the CFTR regulator domain (IPR025837) of the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-06 in 623932 control chromosomes (gnomAD v4.0). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. The Ile840Thr variant has been reported in the literature in a carrier screening study (Picci_2010). These report(s) do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis.
